X-chromosome inactivation and autoimmunity

چکیده
We thank Dr Ozcelik for his careful reading of our review and his thoughtful letter. Dr Ozcelik raises the issue of nonequivalence of monosomy X frequency and X-chromosome inactivation pattern (XCIP). The first refers to the proportion of cells with a single X chromosome, wherein transcription occurs from the lone X chromosome. The second refers to the ratio of inactivated maternal to paternal X chromosome, wherein transcription does not occur from the inactivated chromosome. We agree that although frequency of monosomy X and XCIP are different, both estimate the degree of inequality in X-chromosome dosage. Thus, while direct comparisons may not be possible, it is reasonable to use them as indices for processes inactivating the X chromosome. Dr Ozcelik references 2 unpublished studies refuting XCIP skewing in primary biliary cirrhosis. These studies are unavailable to us and we are unable to comment on them. Finally, our comments about age-related XCIP and the pathogenesis of certain autoimmune diseases have been misinterpreted by Dr Ozcelik. Certainly, many of these autoimmune diseases tend to be more prevalent in older rather than younger patients, and the increased prevalence of XCIP with age suggests there may be some relationship between the two. However, we clearly point out that “no studies have formally linked age related XCIP skewing with the development of autoimmune disease,” and that this topic “deserves further research.”1 Thus, we read with interest Dr Ozcelik’s further thoughts on unequal X-chromosome expression and autoimmune disease.

X Chromosome Inactivation in Opioid Addicted Women
Introduction: X chromosome inactivation (XCI) is a process during which one of the two X chromosomes in female human is silenced leading to equal gene expression with males who have only one X chromosome. Here we have investigated XCI ratio in females with opioid addiction to see whether XCI skewness in women could be a risk factor for opioid addiction. Methods: 30 adult females meeting DS...
متن کاملThe role of X-chromosome inactivation in female predispositionto autoimmunity
We propose that the phenomenon of X-chromosome inactivation in females may constitute a risk factor for loss of T-cell tolerance; specifically that skewed X-chromosome inactivation in the thymus may lead to inadequate thymic deletion. Using a DNA methylation assay, we have examined the X-chromosome inactivation patterns in peripheral blood from normal females (n = 30), female patients with a va...
متن کاملTheory and treatment of the X-inactivation chimera in female-prevalent autoimmune disease.
The vast majority of patients with systemic autoimmune diseases such as Sjögren's syndrome and systemic lupus erythematosus are female. The female prevalence of such autoimmune diseases is poorly understood. In theory, X-chromosome inactivation and resultant tissue chimerism may explain the female predisposition to systemic autoimmunity. For example, autoreactive T cells may fail to be tolerize...
متن کاملThe X chromosome and the sex ratio of autoimmunity.
The number of human conditions that are currently considered to be autoimmune diseases (AID) has been steadily growing over the past decades and it is now estimated that over 10 million people are affected in the United States. One of the major shared features among AID is the predominance in the female sex which in some cases changes with the age at disease diagnosis. Numerous hypotheses have ...
متن کاملX chromosome inactivation patterns correlate with fetal-placental anatomy in monozygotic twin pairs: implications for immune relatedness and concordance for autoimmunity.
BACKGROUND Monozygotic (MZ) twinning is a poorly understood phenomenon that may result in subtle biologic differences between twins, despite their identical inheritance. These differences may in part account for discordant expression of disease in MZ twin pairs. Due to their stochastic nature, differences in X chromosome inactivation patterns are one source of such variation in female MZ twins....
متن کاملIs autoimmunity a matter of sex?
Autoimmune diseases include several conditions that cumulatively are estimated to affect over 5% of the US population with a striking female predominance reported for most of them. The cause and mechanisms of this sex bias remains unknown despite multiple proposed hypotheses. Indeed, it is well established in several experimental settings that the human immune system exhibits sexual dimorphism ...
